ELISPOT全名为酶联免疫斑点检测,英文:Enzyme-linked Immunospot Assay。它结合了细胞培养技术与酶联免疫吸附技术(即ELISA技术),是从单细胞水平检测分泌抗体细胞(ASC) 或分泌细胞因子(CK) 细胞的一项细胞免疫学检测技术。ELISPOT技术原理就是用抗体捕获培养中的细胞分泌的细胞因子,并以酶联斑点显色的方式将其表现出来。由于ELISPOT方法具有较高的特异性和敏感性,易操作,成本相对流式细胞分析术也较低,已被广泛用于分泌CK细胞检测或ASC测定中,对探索自身免疫系统**发病机制具有重要意义。

WhyFluorescentEliSpot?

TheEliSpot assay isknowntobeone of the most sensitive cellularimmunoassaysavailable,allowingthe detection of cytokineproducing T-cells evenat thesingle celllevel. So far, onelimitation in EliSpot technologyhasbeen the lackto detectT-cells which simultaneouslyproducemultiplecytokines.

Toovercomethisproblemresearches have begun to develop EliSpotassays basedonfluorescentlabeling. Detection of multiplecytokinessecretingexpressinglymphocytes such as doubleIFN-γ/IL-2 producingT-cellswith thisnewEliSpot(FluoroSpot)-technology could provideadditionalandnewinformations regarding the clinical outcomeinpatientswithvarious viral infections and other questions.

Fluorescent-based two-color IFN-γ/Il-2-assay interpretedwiththeAIDEliSpot software: The AID reader captures theCy-3/IL-2signal(A)before capturing the FITC/IFN-γ signal(B).Thesoftwareoverlays both images (C) and createsanartificialimagehighlighting the double-stained spots (D).
